Screening of Glycerol-utilized Anaerobic Bacteria for Biobutanol Production

Abstract:

Nowadays, there has been an increasing tendency to increase the production of biodiesel, as a result, the amount of glycerol has been increasing accordingly since it is a by-product of biodiesel production. Glycerol is a good carbon source and it could be used as a substrate for industrial fermentation. This study focus on the screening of anaerobic bacteria which utilized glycerol for butanol production. Soil samples from six agricultural sites were collected from fields of banana, mango, wing bean, rubber, cassava and longan. There are 271 isolates screening from different colony morphology such as color, surface, shape and margin. Even though, merely 133 isolates were gram-positive, rod-shaped and spore-forming. To examine the potential of acetone-butanol-ethanol production, these 133 isolates were cultured in T6 medium utilized glycerol or glucose as a carbon source under anaerobic condition. After HPLC analysis, only 9 isolates could produce butanol from glycerol. The isolates C22, C13 and B21 could produce the highest butanol concentrations of 2.50, 2.37 and 1.76 g/L, respectively. Additionally, the concentrations of acetone and ethanol could be obtained from isolates C22 (0.49 and 4.51 g/L), C13 (0.97 and 3.72 g/L) and B21 (0.42 and 4.63 g/L). Phenotypic characterization by biochemical test was studied and the results showed that each isolate could be motile but couldn’t produce catalase, gelatinase and amylase. All 3 isolates have been able to utilize carbon sources, such as glucose, mannose, cellobiose, lactose, maltose and glycerol. Consequently, the morphology and phenotypic characteristics of each isolate were similar to those of Clostridium sp.
