Abstract:
This work is about the hydrolysis of sucrose into fructose and glucose (inverted syrup) by using invertase [INVERTIN®; (EC.3.2.1.26)]. Its catalytic parameters are as follow: pHoptimum = 5.0, Toptimum = 60oC, Ea = 33 kJ/mol, .H = 30 kJ/mol, KM = 19.6 mM and Vmax = 2.12 U/mL. INVERTIN® was stable for 30h at pH 5.0 and 37oC and its activity was not affected by acetate buffer concentration which was set at 0.01 M. The sucrose hydrolysis was evaluated through a fed-batch process whose sucrose solution (16 g/L) was added according to a decreasing exponential feeding mode. The filling-up times (.) were 2h and 4h and the exponential addition constant values (k) were set at 0.2h-1, 0.5h-1 or 0.8h-1. The yield attained was 100% in all feeding conditions even when the commercial sugar was a substrate. It was also noted that the total reducing sugar formation (TRS) presented distinct profiles as k was varied from 0.2 to 0.8h-1, reflecting different hydrolysis rates (dMTRS/dt) along the reaction; when . = 4h, (dMTRS/dt) was the same for k of 0.5h-1 and 0.8h-1. More tests are thus needed in order to verify whether .>4h may lead to (dMTRS/dt) regardless of k value.
